Nata a Karachi nel 1946 prima della separazione del Pakistan dall'India, Malani fu costretta a rifugiarsi con la famiglia prima a Calcutta e Pune e poi a Mumbai, dove vive e lavora dal 1958. Lì ha realizzato molte opere ed ha conquistato una nuova posizione per le donne artiste, diventando l'icona delle giovani generazioni di artisti indiani. Le sue opere provocatorie sono state esposte in tutto il mondo in importanti personali e collettive tra cui la Biennale di Istanbul (2003) e di Venezia (2005). Robert Storr l'ha scelta anche per la 52.Biennale di Venezia (2007).
