A total of 160 clinical (b…lood, tracheal and cloacal swabs) and post-mortem (brain, lung, colon and spleen) samples were collected from broiler and layer chickens. All the samples were inoculated onto 10-day-old embryonated chicken eggs through allantoic sac route and in the chicken embryo fibroblasts (CEFs) cell culture. The allantoic fluid of the dead embryos and the infected culture fluid of the CEF were harvested at 48 and 96 hours of post-infection, respectively. The HI and RT-PCR were employed to detect NDV in tissue homogenates of all the samples. Inoculum of only cloacal swab and colon showed HA and HI activities. The anti-NDV hyperimmune serum revealed complete inhibition of the 4 haemagglutination unit of each isolate of viruses isolated from broiler and layer chickens. Higher rate of detection of NDV was recorded with RT-PCR assay than HI test. Therefore, RT-PCR can be used for the rapid and confirmatory detection tool of NDV at the field level of Bangladesh. 108 pp. Englisch.

Se emplearon la IH y la RT-PCR para detectar el VEN en los homogeneizados tisulares de todas las muestras. Sólo el inóculo del hisopo cloacal y del colon mostró actividades HA e HI. El suero hiperinmune anti-VND reveló una inhibición completa de las 4 unidades de hemoaglutinación de cada aislado de virus de pollos de engorde y gallinas ponedoras. Se registró una mayor tasa de detección del VEN con el ensayo RT-PCR que con la prueba HI. Tous les échantillons ont été inoculés sur des oeufs de poule embryonnés de 10 jours par la voie du sac allantoïdien et dans la culture cellulaire de fibroblastes d'embryons de poulets (CEFs). Le liquide allantoïdien des embryons morts et le liquide de culture infecté des CEF ont été récoltés respectivement 48 et 96 heures après l'infection. L'IH et la RT-PCR ont été utilisés pour détecter le VND dans les homogénats de tissus de tous les échantillons. Seuls les inoculums de l'écouvillon cloacal et du côlon ont montré des activités HA et HI. Le sérum hyperimmun anti-VND a révélé une inhibition complète des 4 unités d'hémagglutination de chaque isolat de virus isolés de poulets de chair et de poules pondeuses. Le taux de détection de la NDV a été plus élevé avec le test RT-PCR qu'avec le test HI. Par conséquent, la RT-PCR peut être utilisée comme outil de détection rapide et de confirmation du VND sur le terrain au Bangladesh.
