Cytokininen werden ontdekt door F Skoog, C Miller en medewerkers in de jaren vijftig van de vorige eeuw als factoren die de celdeling bevorderen (cytokinese). De eerste ontdekte cytokinine was een adenine (aminopurine) derivaat genaamd 'kinetine' (6-furfuryl aminopurine), dat werd geïsoleerd als een DNA-afbraakproduct. Het eerste bekende natuurlijke cytokinine werd gezuiverd uit onrijpe maïskorrels en kreeg de naam 'zeatine'. Er zijn vandaag de dag nog verschillende andere cytokinines met verwante structuren bekend. Cytokininen zijn aanwezig in alle plantenweefsels. Ze zijn overvloedig aanwezig in de wortelpunt, schiet apex en onrijpe zaden. Hun endogene concentratie ligt in het lage nanomolaire bereik. Typisch zijn verschillende soorten cytokinines en hun gemodificeerde vormen aanwezig in een bepaald weefsel. Cytokininen kunnen over grote afstanden of in de directe omgeving van de cytokinine-producerende cellen werken (paracriene signalering). Cytokininen kunnen ook inwerken op de cel die ze heeft geproduceerd (autocriene signalering). Cytokininen worden ook geproduceerd door cyanobacteriën, sommige plantpathogene bacteriën.
