In der Urologie z.B. kommt es durch Schwäche des äuÃeren Muskelrings und Beckenbodens zur Belastungsinkontinenz (BI) als Spätfolge von traumatischen Geburten, Operationen oder altersabhängiger Degeneration. BI, also Urinverlust aufgrund von Belastung, hat weitreichende Folgen für die Sozioà konomie und Lebensqualität der Betroffenen. In den letzten Jahren versuchte man neue Therapiemethoden zu erforschen und entwickeln, die die Krankheiten nicht nur behandeln, sondern auch heilen kà nnen. Stammzellen aus dem Knochenmark (KMSZ) kà nnen sich unter anderem auch in Muskelzellen differenzieren mit dessen Hilfe man die Muskeln der Harnrà hre wieder regenerieren kà nnte. Das Werk in englischer Sprache beschreibt und diskutiert anhand eines Tiermodells, wie man menschliche KMSZ isolieren, kultivieren, markieren und transplantieren kann und dass sie längere Zeit in artfremdem Gewebe (in Ratten) überleben kà nnen. Die Gewebe wurden zu verschiedenen Zeitpunkten anhand von Immunfärbungen und neuster Mikroskopier Techniken (3D Imaging) untersucht. Das vorliegende Tiermodell zeigt auf faszinierende Weise wie das Potential von Stammzellen sich myogen zu differenzieren und ins Gewebe zu integrieren für Stammzell-basierte Therapien genutzt werden kann. Erstmals konnte die myogene Differenzierung in einer Entwicklungsreihe gezeigt werden. Diese Ergebnisse versprechen neue Mà glichkeiten einer Regeneration der BlasenschlieÃmuskulatur mittels patienteneigener Stammzellen und zukünftig für die Behandlung anderer Muskelerkrankungen.
