Gromov [17], is one of three general methods in immersion-theoretic topology for solving a broad range of problems in geometry and topology. The other methods are: (i) Removal of Singularities, introduced by M. Gromov and Y. Eliashberg [8]; (ii) the covering homotopy method which, following M. Gromov's thesis [16], is also referred to as the method of sheaves. The covering homotopy method is due originally to S. Smale [36] who proved a crucial covering homotopy result in order to solve the classification problem for immersions of spheres in Euclidean space. These general methods are not linearly related in the sense that succes sive methods subsumed the previous methods. The method he used was an ingenious histological technique (La reazione nera) which brought him fame for the discovery of neuronal networks and culminated in the award of the Nobel Prize for Physiology and Medicine in 1906. This technique, however, was not easily reproducible and led to a long-lasting controversy about the reality of the Golgi apparatus. In recent years immunochemical techniques and molecular cloning approaches opened up new avenues and led to an ongoing resurgence of interest. Instead, inflammatory indices are emerging as characteristic features of the pathology and pathophysiology of CB/COAD. It is possible that continuous inflammatory processes cause structural changes and thus provide the anatomical basis for airway obstruction which, once estab lished, is only partly reversible. We feIt it was timely to gather leading international researchers who could provide critical updates on CB/COAD, its pathology and clinical/ experimental assessments. All of them have subsequently been shown to have significant and specific biological activities not only in inflammatory responses but also in other physiological pro cesses. It was our aim to complete the book in advance of the lectures to be given at the 'International Symposium on Asthma and Allergic Diseases in Childhood', in Berne, Switzerland, on March 25th-27th, 1993. This symposium commemorates the 10th anniversary of the Swiss Association of Parents of Asthmatic and Allergic Children. The incidence of asthma and allergies generally is on the increase and demands everyone's attention, including that of physicians, researchers, allied health professionals and parents. There have been no recent breakthroughs in our understanding of asthma pathogenesis or treatment. However, forms of treatment, pathogenetic insights and public awareness are undergoing rapid changes as our knowledge of atopic diseases increases. For this reason we chose the title 'Update on Childhood Asthma'. The purpose of the papers is to summarize recent research results, to provide an update on the current approach to therapeutic interventions and to alert readers to the psychosomatic aspects of asthma. Whether the state of knowledge has already changed while you are holding this book in your hands is not so much dependent on a delay in publishing but on the speed of appearance of new research results. Free radicals form in biological systems through metabolism, but it is also realized that exogenous environmental sources, such as radiation, food, and drugs, contribute significantly to the generation of free radicals in biological systems. Being reactive species, free radicals are short-lived and do not travel far from cellular targets. Their concentration in biological systems is very low and is difficult to detect directly by electron spin resonance spectroscopy (ESR). Indirect methods of reactions of radicals with specific biomolecules are also sufficiently sensitive to detect quantitatively their presence. Thus the response of antioxidant defenses which react with radical species, can serve as an indirect measure that free radicals have been formed. Redox-based antioxidants change their oxidation state and antioxidants become free radicals themselves. Often, however, the antioxidants give rise to more persistent free radicals, sometimes owing to delocalization of the lone electron around ring structures (in vitamin E, ubiquinones, and certain carotenes). Persistent free radicals react only rarely and the precursors often can be regenerated in biological systems. In recent years, it is becoming clearer from biochemical studies on how the major lipophilic antioxidants work. Particular attention has been given to vitamin E and quinones found in animal and plant membranes and in carotenoids, for the protection of membranes in lipoprotein systems.
