Since 1983, the metabolic defect in some 20 different peroxisomal disorders has been described. The best known conditions include Zellweger syndrome, rhizomelic chondrodysplasia punctata and X-linked adrenoleukodystrophy and, in the most recent edition of The Metabolic and Molecular Basis Inherited Disease, edited by Scriver and colleagues, more than 100 pages are now devoted to the subject. Progress in our understanding of these conditions, and their diagnosis, results from the application of a variety of laboratory investigations. These include microscopic studies, analysis of metabolites (very long-chain fatty acids, bile acids, and plasmalogens), enzyme studies (peroxisomal beta-oxidation pathway and dihydroxyacetone phosphate acyltransferase), immunodetection of peroxisomal (membrane) proteins and molecular analysis of mutant DNA. In order to encourage a greater awareness in this field and the diagnostic protocols required, an international course was organised in Gent, Belgium, in May 1994, on the clinical and biochemical diagnosis of peroxisomal disorders. A number of international experts in the field who provided intensive hands-on experience over 3.5 days, have now collected their course work and reviews together in this Handbook. The volume is introduced by Sidney Goldfischer, who in 1973 was the first to recognise the absence of peroxisomes in Zellweger syndrome, but whose observations were not fully appreciated for a further decade. This handbook provides the most comprehensive and detailed account of laboratory methods for the diagnosis of peroxisomal disorders. The methods are clearly presented and well illustrated, and should allow laboratories to introduce these methods into their repertoire. It naturally prolongs the theme of the monograph 'The Geometry of La grange spaces: Theory and Applications', written together with M. Anastasiei and published by Kluwer Academic Publishers in 1994. The existence of Lagrangians of order k 1 has been contemplated by mechanicists and physicists for a long time. Einstein had grasped their presence in connection with the Brownian motion. They are also present in relativistic theories based on metrics which depend on speeds and accelerations of particles or in the Hamiltonian formulation of non linear systems given by Korteweg-de Vries equations. There resulted from here the methods to be adopted in their theoretical treatment. One is based on the variational problem involving the integral action of the Lagrangian. A second one is derived from the axioms of Analytical Mechanics involving the Poincare-Cartan forms. The geometrical methods based on the study of the geometries of higher order could invigorate the whole theory. Efficient immunosuppressive regimens have been developed with the objective of very good results at 1, 3 and 5 years. Successful transplants, however, are significantly less frequent at 10 and 20 years, and many patients require retransplantation. Many factors are involved in late graft loss and it is now well recognized that, in addition to chronic rejection, a number of non-immunologic factors play a prominent role. In the case of renal transplantation, a reduced mass loss (transplantation of a single kidney, sometimes from an aged donor, ischemic injury and alteration of some nephrons in the case of early acute rejection) will result in slowly progressing chronic renal failure, even in the absence of any supplementary attack of an immunological nature. The new treatments must be analyzed in the light of their capacity to reduce these late failures. While Pau now exists largely on agriculture and, more recently, oil extraction and refining, a century ago it was a winter resort that rivalled Cannes and Nice; indeed, Baedeker judged the air of Pau to be preferable for those of a nervous disposition. Malthusian population predictions continue to alarm agricultural researchers, especially plant breeders, to seek new technologies that will continue to allow us to produce more and better food by fewer people on less land. Both improvement of existing cultivars and development of new high-yielding cultivars are common goals for breeders of all crops. In vitro haploid production is among the new technologies that show great promise toward the goal of increasing crop yields by making similar germplasm available for many crops that was used to implement one of the greatest plant breeding success stories of this century, i. e. , the development of hybrid maize by crosses of inbred lines. One of the main applications of anther culture has been to produce diploid homozygous pure lines in a single generation, thus saving many generations of backcrossing to reach homozygosity by traditional means or in crops where self-pollination is not possible. Because doubled haploids are equivalent to inbred lines, their value has been appreciated by plant breeders for decades. The second international symposium, Systems Approaches for Agricultural Development, held in Los Baños, 6-8 December 1995, fostered this link between the bio-physical sciences and the social sciences in the choice of keynote papers and oral presentations, a selection of which is included in this book. The book's contents further reflect how systems approaches have definitely moved beyond the research mode into the application mode. The large number and high quality of interdisciplinary research projects reported from different parts of the globe, to determine land use options that will meet multiple goals and yet sustain natural resource bases, is a key indicator of this `coming of age'.
