However, such regulation is only one of many factors that contribute to the levels of a given p- tein. One major factor that has been relatively obscure until recently has been the contribution of protein degradation to the regulation of the steady state level of protein expression and protein function. This rapidly evolving field has made a significant mark on the scientific community, as highlighted by the Award of the Nobel Prize in Chemistry for 2004 to Aaron Ciechanover, Avram Hershko and Irwin Rose for their pioneering work on the ubiquitin-proteasome system (UPS) of protein degradation, which is the subject of this volume. In recent years e- dence has been accumulating that suggests a role for UPS function in both ph- iological and pathological settings. In particular, studies have implicated a central role for the UPS in cell cycle regulation, cancer and neurodegeneration. Prior to Banting and Best, chnicians were unaware of the ravages of diabetes, patients simply wasted away and died. Following the purifica tion of insulin, clinicians now had to deal with diabetic retinopathy, diabetic neph ropathy and all the other complications of long-term diabetes. A little over 50 years ago, the first successful human kidney transplant was performed in Boston. The first 30 years of the experience had successes when compared to the alternative but were a constant struggle to get even 50% of the grafts from deceased donors to survive more than a year. However, the science continued to advance knowledge of the immune response. With this came more and increasingly powerful tools for the clinician. Suddenly, success rates of 80-90% at one year were attainable. With this success came new problems, new complications and clinicians now had to worry about the long-term consequences of their therapy as patients were surviving with functional grafts for extended periods. A particular infectious complication evolved with the application of ever more powerful immunosuppressant drugs. Astute clinicians noted that occasionally cellular rejections seemed to get worse with steroids. This may seem unusually large, but I include in 'verification' all debugging and correctness checking activities, not just writing and running testbenches. Every time a hardware designer pulls up a waveform viewer, he or she performs a verification task. With today's ASIC and FPGA sizes and geometries, getting a design to fit and run at speed is no longer the main challenge. It is to get the right design, working as intended, at the right time. Unlike synthesizable coding, there is no particular coding style nor language required for verification. The freedom of using any l- guage that can be interfaced to a simulator and of using any features of that language has produced a wide array of techniques and approaches to verification. The continued absence of constraints and historical shortage of available expertise in verification, c- pled with an apparent under-appreciation of and under-investment in the verification function, has resulted in several different ad hoc approaches.
