L'enzyme a ete partiellement purifiee par precipitation au sulfate d'ammonium avec une purification de 4,84 fois et un rendement de 7,42 %. L'enzyme a montre une activite optimale a une temperature de 60 0C et un pH de 10. The demand for microbial enzymes in industrial fields is increasing day by day due to their applications in clean and cost effective biotechnological processes. A halo-tolerant and alkaliphilic bacterium, Kh-30-2, was isolated from saline soil of the desert of Kutch, Gujarat. The organism secreted a range of extracellular enzymes including protease, cellulase and lipase with 10% NaCl and pH 9. It secreted alkaline protease optimally with 0.5 % w/v, peptone and yeast extract. The enzyme was partially purified by ammonium sulphate precipitation with 4.84 fold purification and 7.42% yield. The enzyme displayed optimum activity at 60°C temperature and pH 10. Now a day, majority of commercial cellulases are being produced by fungi such as, Trichoderma. However halophilic fungi can be a valuable resource for the same. The present study deals with the extracellular cellulases from Halophilic fungi isolated from the Great Rann of Kutch, India. The isolates, designated as B10-1 and D10-2 were having mycelia structure similar to Aspergillus spp. and grew optimally with 10% w/v NaCl. B10-1 and D10-2 grew optimally at pH 6 with slow growth at higher pH. In case of B10-1 and D10-2, the optimum cellulose concentration for cellulase production was 0.5% w/v and 1.5% w/v respectively. Both the isolates produced cellulase optimally at 10% NaCl and pH 6. B10-1 and D10-2 exoglucanase were purified to 10.98 fold and 4.23 fold respectively by (NH4)2SO4 fractionation. Partially purified extracellular exoglucanase was characterized further. The enzymes displayed optimum activity at 60°C beyond which the activity gradually declined. The enzymes were active at 60°C for more than an hour. The enzymes of both the isolates withstand even at 70°C for more than half an hour. 76 pp. Englisch.

Actinomycetes are a family of bacteria that produce over two thirds of the clinically useful antibiotics of natural origin. The present study deals with production and extraction of antimicrobial metabolites from haloalkaliphilic actinomycetes from the Gulf of Khambhat situated in the Bhavnagar District of the Gujarat. Out of 40 isolates, isolate GhM-HA-6 was found to secrete potential antimicrobial metabolites against many Gram-positive bacteria like M.leuteus, S.flexneri, S.aureus and S.epidermis. It produced the antibiotic optimally during stationary phase with pH 9 and 10% w/v NaCl in starch broth. The crude antimicrobial substance was isolated by using various solvent systems but giving the best separation with chloroform and methanol-ethyl acetate. The study reveals that the haloalkaliphilic actinomycete GhM-HA-6 produces broad spectrum of antimicrobial metabolites which can be exploited for biotechnological potential and improve as promising sources for new antimicrobial compound. 60 pp. Englisch.

There is an alarming scarcity of new antibiotics currently under development in the pharmaceutical industry. The present study highlights the isolation and characterization of haloalkaliphilic actinomycetes with respect to their antimicrobial potential. A total of 14 halophilic, alkaliphilic and haloalkaliphilic actinomycete isolates were isolated from the saline soil samples collected from the saline desert of Kutch under varied conditions of salt (0-20% w/v NaCl) and pH (7-10). The organisms belonged to the genus Streptomyces with fluffy filamentous aerial mycelia. Their ability to secrete antimicrobial metabolites was tested against a range of Gram-positive and Gram-negative test cultures. Three isolates H3, H4 (Halophilic) and HA5 (haloalkaliphilic) emerged as the potent antimicrobial producers. They produced the antimicrobial agent against Micrococcus in liquid culture after 15 days of incubation under shaking conditions.
