This study explored the application of tetracycline HCl (Tc HCl) films based on gel forming polymers as drug release regulators for local treatment of periodontal pockets. Different film formulations were simply prepared by 'solvent casting technique'. Films were suitable for intra-pocket application, they were hard enough to be easily inserted into the periodontal pocket. Immediately after application, they hydrate by the action of gingival crevicular fluid forming a bioadhesive gel which allow the formulation to get access to the entire pocket. Films exhibited a sustained release pattern. The stability study for the selected formula (Tc HCl/Na CMC) indicated that the five months storage period did not significantly influence the drug release behavior or its antibacterial activity. Clinical evaluation indicated that the film was well tolerated, no signs of irritation or prolonged foreign body sensation were reported by the test subjects and significant reduction in the pocket depth was observed. 112 pp. Englisch.…

Cette étude a exploré l'application de films de chlorhydrate de tétracycline (Tc HCl) à base de polymères gélifiants comme régulateurs de libération du médicament pour le traitement local des poches parodontales. Différentes formulations de films ont été préparées simplement par la « technique de coulée en solvant ». Les films étaient adaptés à une application intra-poche, ils étaient suffisamment durs pour être facilement insérés dans la poche parodontale. Immédiatement après l'application, ils s'hydratent sous l'action du liquide gingivo-creviculaire, formant un gel bioadhésif qui permet à la formulation d'atteindre l'ensemble de la poche. Les films ont présenté un profil de libération prolongée. L'étude de stabilité de la formulation sélectionnée (Tc HCl/Na CMC) a indiqué que la période de stockage de cinq mois n'avait pas d'influence significative sur le comportement de libération du médicament ou son activité antibactérienne. Este estudo explorou a aplicacao de filmes de cloridrato de tetraciclina (Tc HCl) a base de polimeros gelificantes como reguladores de libertacao de farmacos para o tratamento local de bolsas periodontais. Diferentes formulacoes de filmes foram preparadas de forma simples atraves da tecnica de moldagem por solvente. Os filmes eram adequados para aplicacao intra-bolsa, sendo suficientemente rigidos para serem facilmente inseridos na bolsa periodontal. Imediatamente apos a aplicacao, hidratam-se pela acao do fluido crevicular gengival, formando um gel bioadesivo que permite que a formulacao aceda a toda a bolsa. Os filmes exibiram um padrao de libertacao sustentada. O estudo de estabilidade para a formula selecionada (Tc HCl/Na CMC) indicou que o periodo de armazenamento de cinco meses nao influenciou significativamente o comportamento de libertacao do farmaco ou a sua atividade antibacteriana.
